Taq DNA Polymerase Antibody is a monoclonal antibody that binds Taq polymerase and inhibits its activity until reaction temperature is elevated. At that point, the Taq Antibody is denatured and releases its hold on Taq polymerase, allowing DNA synthesis to proceed. The binding of the Taq Antibody to Taq polymerase is the premise of hot-start PCR.
By preventing Taq polymerase activity until the denaturation step of PCR is underway, nonspecific amplification due to mispriming and/or formation of primer dimers during PCR assembly is prevented. As a result, experimental background is significantly decreased, and reaction specificity is increased.
Taq Antibody can be added to Taq DNA Polymerase or Multiplex PCR before PCR to lower background, increase specificity, and allow room temperature reaction assembly.
Applications
- Hot-Start PCR
- Multiplex PCR
- Specialty PCR
- PCR
- Routine PCR
Antibody performance
Following incubation of Taq antibody and Taq polymerase at 25°C for 10 min, inhibition of Taq polymerase activity was confirmed as greater than 90% (at 55°C for 10 min).
Following heat treatment at 94°C for 30 seconds, Taq antibody-bound Taq polymerase was confirmed to recover almost 100% activity (at 55°C for 10 minutes).
